font-family:"Arial","sans-serif";color:#95B3D7'>How motor proteins effect endosome transport and fission is investigated through quantitative biophysical measurements<i>. </i>Opposing teams of Kinesin and Dynein are shown to engage in a molecular tug-of-war on single endosomes. Surprisingly, many (4 to 8) weak and detachment-prone dyneins are in tug-of-war against a single strong and tenacious kinesin. We show how this clever choice of opposing motor-teams allows efficient minus-end transport <i>(required for endosome maturation)</i> concurrently with controlled fission of endosomes <i>(required for receptor recycling)</i>. This appears to be the first demonstration that the two major microtubule motors (Kinesin and Dynein) function differently at the molecular level inside cells, and of how this difference may regulate an important cellular process, namely endosome biogenesis.</span></p> <p class=MsoNormal style='margin-top:0cm;margin-right:3.6pt;margin-bottom:0cm; margin-left:63.0pt;margin-bottom:.0001pt'><span style='font-family:"Courier New"; color:silver'>&nbsp;</span></p> <p class=MsoNormal style='margin-top:0cm;margin-right:3.6pt;margin-bottom:0cm; margin-left:36.0pt;margin-bottom:.0001pt'><span style='font-family:"Arial","sans-serif"; color:silver'>V Soppina, A K Rai and R Mallik</span></p> <p class=MsoNormal style='margin-top:0cm;margin-right:3.6pt;margin-bottom:0cm; margin-left:36.0pt;margin-bottom:.0001pt'><i><span style='font-family:"Arial","sans-serif"; color:silver'>Simple non-fluorescent polarity labeling of Microtubules for Molecular Motor assays</span></i></p> <p class=MsoNormal style='margin-top:0cm;margin-right:3.6pt;margin-bottom:0cm; margin-left:36.0pt;margin-bottom:.0001pt'><span style='font-family:"Arial","sans-serif"; color:silver'>Biotechniques <b>46</b>, 297 (2009) </span><span style='font-family:"Arial","sans-serif";color:#CC99FF'><a href="http://www.tifr.res.in/~roop/Publications_files/MT-Polarity-Labelling-Soppina2009.pdf"><span style='color:#CC99FF'>PDF</span></a> <u><a href="http://www.ncbi.nlm.nih.gov/pubmed/19594454"><span style='color:#CC99FF'>Abstract</span></a></u></span></p> <p class=MsoNormal style='margin-top:0cm;margin-right:58.45pt;margin-bottom: 0cm;margin-left:63.0pt;margin-bottom:.0001pt'><span style='font-size:9.0pt; font-family:"Arial","sans-serif";color:#95B3D7'>A new method to identify the polarity (minus and plus) of in vitro assembled microtubules.
